Given that Pejac used the words "focus and suspense" to describe making the painting at night, and assuming that this is a truthful description, I would say that it is unlikely, though obviously not impossible, that he put down the base layer of paint on the same night. I do agree that there probably was graffiti on the door beforehand, though.
When considering your observation in a previous post that it is not surprising that Pejac's artwork got painted over, I think it is worth bearing in mind that it had been in place since at least 29 June 2015 (going by the time the work was first posted on Instagram by the artist).
If the throw-up that went over was painted around the time it was posted by sho_altmed on Instagram (1 February 2016), Pejac's piece lasted about 7 months.
On 18 October 2015, Pejac made the below repost of an image by keystonowhere which shows his piece with a newer throw-up above it. The original photo was also posted on 18 October. If this photo was taken around the time it was posted, we then know that Pejac's piece was gone over sometime between mid-October 2015 and 1 February 2016.
